Openly gay characters
- Patty Bouvier - She almost married Veronica in "There's Something About Marrying". She may be bisexual as she had a relationship with Principal Skinner in "Principal Charming".

Hinted to be Gay
- Waylon Smithers - Various scenes in the series hint he is gay and in love with Mr. Burns. Also once had a relationship with John.
- Duffman - Hinted to be
- Lenny Leonard - Sometimes hinted as being attracted to Carl.
- Carl Carlson - Sometimes hinted as being attracted to Lenny.
- Santa's Little Helper - Intrigued by male dogs at the Springfield Gay Pride Parade
- Edna Krabappel - Hinted in Rome-Old and Julie-Eh that she may be in a relationship with Patty Bouvier.
Transsexual
- Judge Constance Harm indicates that she was born a man
- Brunella Pommelhorst says she will return next semester as Mr. Pommelhorst, the new shop teacher.
