- Homer: Homer is one of the playable characters, he can jump, kick, stun enemies using his burp, turn into Homer Ball, Helium Homer, Gummi-Homer and (only on Xbox 360/PS3 version) "Lava-Homer".
- Bart: Bart is one of the playable characters, he can jump, kick, use his slingshot, turn into bart to glide, climb walls, use ziplines, and grapple onto objects. He can also turn into "Robo-Bart" (Xbox 360/PS3 version only).
- Matt Groening: he is the main boss in the last part of the level. He changes positions and draws sketches of Bender and Zoidberg so they become real and attack Homer and Bart.
- Blue-haired lawyers: enemies throughout the level, except in the last part. They attack by launching papers through their briefcases.
- Bender Bending Rodriguez: one of the main characters of Futurama. Multiple Benders are drawn by Matt Groening as enemies in the last part of the level to attack Homer and Bart by throwing beer bottles.
- Zoidberg: one of the main characters of Futurama. Multiple Zoidbergs are drawn by Matt Groening as enemies in the last part of the level to attack Homer and Bart by shooting ink at them (Wii/PS2/PSP version only).
