Homer's colonoscopy is performed at the Mayo Clinic, which is famous for its research, educational programs and treatment of difficult cases.
When the assistant uncovers Homer's posterior, the doctor waxes euphoric, comparing it to the Sistine Chapel and to Moby Dick.
While performing the colonoscopy, the doctor finds a CD of the Beatles album Sgt. Pepper's Lonely Hearts Club Band.
When Homer wakes up, he sings the "Hallelujah Chorus" from Handel's Messiah.

When Homer asks Marge if she's getting a breast enlargement, he uses the word "embiggen", one of the show's earliest and most famous neologisms.
Homer uses his "Mmm..." catchphrase when the doctor welcomes him to the Mayo Clinic.
Mr. Burns visits the Mayo Clinic in "The Mansion Family". He goes in for a checkup and is diagnosed with "Three Stooges Syndrome".

Immediately after the colonoscopy, Homer heads for Krusty Burger, in his haste running out of the room in his hospital gown and with the camera still inside him. In real life, because of the preparatory fasting required, it is not unusual for colonoscopy patients to be ravenously hungry following the procedure.
